E-mail: [email protected] Visceral leishmaniasis is an emergent zoonosis with an increasing number of new cases in Brazil where the domestic dog is an important parasite reservoir in the infectious cycle of Leishmania chagasi. An enzyme-linked immunosorbent assay (ELISA), based upon the use of a total soluble antigenic preparation of L. chagasi, was adapted for the detection of IgM antibodies in the serum of infected dogs. Optimal dilutions of the antigen, using positive and negative reference sera, were determined by checkboard titrations. The specificity and sensitivity of the ELISA were 100 %. A total of 110 serum samples were taken from dogs in Belo Horizonte, Minas Gerais, Brazil, and examined for anti-L. chagasi IgM antibody by ELISA and indirect fluorescent antibody test (IFAT). About 25% (n=27) of all the dogs tested were found serologically positive for L. chagasi by IFAT, while 89.09% (n=98) were seropositive by ELISA. The results obtained by ELISA and IFAT were significantly different (P<0.01). The combined use of ELISA and IFAT is recommended in order to enable veterinary services to more efficiently detect canine visceral leishmaniasis.

Designing an enzyme-linked immunosorbent assay for detection of anti-penicillin antibodies levels in various species of animals in Khuzestan province, Iran

The penicillin allergy is being increasingly recognized as a significant public health problem. Immunological responses to penicillin and other beta-lactam antibiotics are classified as immediate and non-immediate responses. This research aimed to develop an enzyme-linked immunosorbent assay (ELISA) for the detection of the reactive antibody value against penicillin in various species of animals...

Enzyme-Linked Immunosorbent Assay of Progesterone in Serum Using Penicillinase as Label

An enzyme-linked immunosorbent assay (ELISA) for progesterone measurement in serum or plasma samples using penicillinase as label enzyme is reported. A C3 and C11 derivatives of progesterone were prepared and conjugated to bovin serum albumin (BSA). Polyclonal antibody against these two immunogens were prepared in New Zealand white rabbits. Purified Ig fractions of antibodies were immobilized o...

Adaptation of an indirect enzyme-linked immunosorbent assay by purified gp51SU for detection of antibodies to bovine leukemia virus

The objective of this study was to compare an indirect ELISA, based on a purified 60 kDa envelope glycoprotein (gp51SU), with a Pourquire indirect ELISA for the detection of antibodies to the bovine leukemia virus. For conducting this research, 340 serum samples were collected from two different breeds of cows (Sarabi and Holestin) in different herds. Commercial ELISA revealed positive results ...

Development of an Indirect Enzyme-linked Immunosorbent Assay to Detect Antibodies against Serotype A2013 of Foot and Mouth Disease Virus in Cattle

Foot and mouth disease (FMD) is a contagious animal disease that causes irreparable damage to the economy of countries, including Iran in which this disease is a native one. Among the ways to combat FMD are vaccination and slaughter. Due to the specific situation of Iran, it is not possible to kill infected animals. Therefore, vaccination is the most important way to fight this disease. S...

Validation of a β-ME ELISA for Detection of Anti Leishmania donovani Antibodies in Eastern Sudan

Background: A β-mercaptoethnol (β-ME)-treated promastigote antigen of L. donovani was successfully employed in direct agglutination test (DAT) for the diagnosis of visceral leishmaniasis (VL). Objective: The β-ME-treated antigen was further incorporated into an enzyme-linked immunosorbent assay set-up (β-ME ELISA) and evaluated for VL diagnosis against outcome of reference freeze-dried DAT (FD-...
